Anti-AChR antibodies are detectable by a radiobinding assay. Specimens and references are incubated for one hour with detergent-solubilized fetal and adult AChRs after that hour I125-alpha-bungarotoxin is added to the tubes, and acetylcholine blocking antibodies directed at the acetylcholine binding cite compete with the I125-alpha-bungarotoxin for a limited number of binding sites on the receptors. The resulting acetylcholine receptor-blocking antibody complexes are then precipitated with Con-A Sepharose. After centrifugation, the supernatant is aspirated and the pellet containing AchR/antibody-bound complex is counted in angamma counter. Counts are inversely proportional to the amount of antibodies present.
