The Total T4 Coated Tube RIA Kit is a radioimmunoassay used for determination of total thyroxine in plasma or serum.
Undiluted samples along with standards and controls are incubated with tracer for 2 hours in thyroxine antibody coated tubes. During incubation the thyroxine from the samples, standards, and controls compete for binding with the thyroxine from the tracer to the antibody on the surface of the tubes in the presence of blocking agents for thyroid hormone binding proteins. After incubation, the tubes are washed with water to remove unbound thyroxine and tracer. The tubes are read on a gamma counter following wash steps. The level of thyroxine in the samples, standards, and controls are inversely related to the level of radioactivity that is measured.
