INTENDED USE

The IVD Technologies Acetylcholine Receptor Blocking Antibody Kit is a radioimmunoassay for the semi-quantitative determination of acetylcholine receptor (AChR) blocking antibodies. The measurement of AChR blocking antibodies is useful as an aid in the differential diagnosis of Myasthenia Gravis (MG) in conjunction with other clinical and laboratory findings (see References below).

 

TEST PRINCIPLE

Samples are incubated for one hour at room temperature with detergent-solubilized AChR. During incubation AChR blocking antibody in the sample are allowed to bind with AChR.  After incubation, 125I-?-bungarotoxin is allowed to bind to the remaining binding sites on the AChR. The AChR is then precipitated with Sepharose-Con A. Following centrifugation, the pellet contains 125I-?-bungarotoxin. The amount of 125I-?-bungarotoxin is inversely proportional to the concentration of AChR blocking antibody in the original sample. The percent inhibition of 125I-?-bungarotoxin binding reflects the concentration of these antibodies in the serum sample.
